GLP-2 Isn't a Weight-Loss Peptide: Here's What It Actually Is
Once you know GLP-1 as shorthand for semaglutide, tirzepatide, and the rest of that category, it’s a natural guess that “GLP-2” is just the next one in the series: a newer, stronger version aimed at the same thing. It’s not. GLP-2 is a real, distinct hormone, but it does something almost unrelated to appetite or weight loss.
Same origin, different job
GLP-1 and GLP-2 are actually released together: both come from the same precursor protein (proglucagon) and are secreted by the same intestinal L-cells in response to eating. That shared origin is probably the whole reason for the naming mix-up. But once released, they act on completely different receptors and do completely different things.
GLP-1’s job, covered in more detail in our GLP-1 explainer, is appetite and blood sugar regulation: slowing digestion, signaling fullness, and supporting insulin release. GLP-2’s job is maintaining and growing the lining of the intestine itself. It increases nutrient and fluid absorption, promotes growth of the intestinal mucosa, and reduces gut permeability. It’s not an appetite signal at all.
Where GLP-2 is actually used
The clinical application for GLP-2 is short bowel syndrome, a condition where someone has lost a significant portion of their intestine (often from surgery) and struggles to absorb enough nutrients and fluids from food. A GLP-2 analog called teduglutide is an FDA-approved prescription treatment for exactly that, marketed as Gattex. The goal is the opposite direction from a GLP-1 weight-loss drug: helping the gut absorb more, not less.
The takeaway
If you came across “GLP-2” expecting a weight-loss compound, that’s a reasonable mix-up given the naming, but it’s not what the hormone does. GLP-1 and its receptor agonists (semaglutide, tirzepatide, retatrutide) are the appetite and metabolic category; GLP-2 is a structural-gut-health hormone with an unrelated clinical use.
